The Doctoral Thesis advances recombinant Pichia pastoris (Komagataella phaffii) fermentation engineering by integrating real-time monitoring, hybrid modelling, and predictive control aligned with Industry 4.0 principles. Using HBcAg, LegH, and Qβ producing strains, key fed-batch challenges were addressed: sensors were validated, data processing methods developed, and the prediction capabilities of mechanistic, data-driven, and hybrid models with transfer learning were compared. PI- and MPC-based control was implemented to ensure stable and robust process operation.
